Procedure

283(b) The partially purified 1,1-dimethylethyl (2S)-4-[(3′-{[({3-[({[1,6-diethyl-4-(tetrahydro-2H-pyran-4-ylamino)-1H-pyrazolo[3,4-b]pyridin-5-yl]methyl}amino)carbonyl]-phenyl}carbonyl)amino]methyl}-2′-fluoro-3-biphenylyl)methyl]-2-methyl-1-piperazinecarboxylate was mixed with THF (1 mL) and TFA (0.2 mL). The mixture was stirred at room temperature for 1 h. Purification was done with a Gilson HPLC using TFA conditions. The desired fraction was collected, concentrated and re-dissolved in MeOH. Filtration though an amine cartridge and solvent concentration then afforded the above named product (10 mg). LC-MS m/z 747 (M+H)+.
