To a solution of 4-chloro-N-(1-methyl-1H-pyrazol-4-yl)-7H-pyrrolo[2,3-d]pyrimidin-2-amine (2 g, 8 mmol) in DMF (35 mL) was added N-chlorosuccinimide (1.28 g, 9.6 mmol). The reaction mixture was stirred at 50° C. overnight. Water (50 mL) was added and the solution was extracted with EtOAc (three×25 mL) and the combined organic layers were dried over Na2SO4, and concentrated to dryness. The residue was purified by preparative HPLC to afford the title compound (0.5 g, 22% yield) as a gray solid. 1H NMR (400 MHz, DMSO-d6) δ ppm 11.99 (brs, 1H), 9.62 (brs, 1H), 7.88 (s, 1H), 7.52 (s, 1H), 7.38 (s, 1H), 3.35 (s, 3H). m/z for C10H8Cl2N6: 283.0 (M+H)+.
