To a solution of 3-(3-(tert-butyldimethylsilyloxy)propyl)-6-chloropyrido[3,4-d]pyrimidine-2,4(1H,3H)-dione (3.3 g, 0.0089 mol), K2CO3 (2.468 g, 0.0178 mol) in acetone (10 mL) was added iodomethane (1.51 g, 0.01 mmol). The reaction was stirred at RT for 3 h then diluted with EA (20 mL) and water (5 mL). The organic layer was washed with brine (20 mL), dried over Na2SO4 and concentrated to give 3-(3-(tert-butyldimethylsilyloxy)propyl)-6-chloro-1-methyl pyrido[3,4-d]pyrimidine-2,4(1H,3H)-dione (3 g, 87% yield) as a white solid. MH+ 384, TR=2.166 min. Used without further purification.
