A mixture of N-[(3-bromo-4-fluorophenyl)methyl]-N′-{[1,6-diethyl-4-(tetrahydro-2H-pyran-4-ylamino)-1H-pyrazolo[3,4-b]pyridin-5-yl]methyl}pentanediamide (0.92 g, 1.5 mmol), (3-formylphenyl)boronic acid (292.5 mg crude, 1.95 mmol), Na2CO3 (477 mg, 4.5 mmol) in dioxane/water (3:1, 16 mL) was swept with N2 for about 5 min before Pd(PPh3)4 (87 mg, 0.075 mmol) was added. The tube was sealed and irradiated with a microwave reactor for 30 min at 150° C. The same reaction was then repeated at the same scale. The combined mixture was mixed with water (50 mL), washed with EtOAc/Diethyl Ether (1:1, 50 mL, 2×). Purification via a CombiFlash system (EtAc/MeOH/DCM) then provided the desired product (1.3 g in total from two trials). LC-MS m/z 630 (M+H)+.
