A mixture of 8-chloro-1-(piperidin-4-yl)-1H-[1,2,3]triazolo[4,5-c][1,5]naphthyridine (100 mg, 0.35 mmol), 2-chloro-N,N-dimethylacetamide (46 mg, 0.38 mmol), and K2CO3 (97 mg, 1.04 mmol) in DMF (5 mL) was stirred at r.t. overnight. The solvent was removed, and the residue was extracted with EtOAc (3×10 mL). The combined organic layers were dried over Na2SO4, and concentrated to afford crude 2-(4-(8-chloro-1H-[1,2,3]triazolo[4,5-c][1,5]naphthyridin-1-yl)piperidin-1-yl)-N,N-dimethylacetamide, which was used in the next step without further purification.
