Isopropylmagnesium chloride-lithium chloride complex (1.3 M in THF, 18.3 mL, 23.7 mmol) was added dropwise to a cold (0° C.) solution of 5-bromo-2,4-dimethoxypyrimidine (4 g, 18.3 mmol) in THF (50 mL), under argon. The reaction mixture was allowed to warm to rt and stirred for 1 h at this temperature. Di-tertbutyl azodicarboxylate (4.2 g, 18.3 mmol) was added portionwise. The reaction mixture was stirred for 1 h at rt, diluted with brine, and extracted with EtOAc. The organic layers were combined, washed with brine, dried (Na2SO4), and concentrated. The residue was purified by silica gel column chromatography (hexane/EtOAc, 9:1→7.3) to provide 6.78 g of the title compound. tR: 1.08 min (LC-MS 4); ESI-MS: 371.3 [M+H]+ (LC-MS 4); Rf=0.55 (hexane/EtOAc, 1:1).
