Procedure

To a solution of methyl 2-(4-butyl-2,3-dioxopiperazin-1-yl)-3-phenylpropanoate from Step A (130 mg, 0.39 mmol) in 10 mL THF and 3.9 mL water was added lithium hydroxide monohydrate (50 mg, 1.2 mmol). The reaction was stirred at rt for 2 h and HCl (1M, 1.2 mL) was added. Solvent evaporation gave 2-(4-butyl-2,3-dioxopiperazin-1-yl)-3-phenylpropanoic acid. 1H NMR (CD3OD) δ 7.26 (m, 5H), 5.18 (X of ABX, 1H, J=11.6 Hz, J=5 Hz), 3.55-3.17 (m, 8H), 1.50 (m, 2H), 1.27 (m, 2H), 0.92 (t, 3H, J=7.3 Hz).
