A mixture of 7-iodo-5-nitro-1-benzofuran (1.00 g, 3.46 mmol), 4-aminopyridine (0.39 g, 4.15 mmol), Xantphos (0.20 g, 0.36 mmol), Pd2(dba)3 (0.08 g, 0.09 mmol), NaOtBu (0.47 g, 4.84 mmol) in xylene (200 mL) was heated at 120° C. overnight. The reaction mixture was filtered through Celite and a yellow precipitate was formed which was collected by filtration to yield 0.52 g (60%) of the title product. HPLC 96%, RT: 1.344 (System A; 10-97% MeCN over 3 min). 1H NMR (270 MHz, methanol-d4) δ ppm 6.91-7.12 (m, 4H) 8.00 (d, J=2.23 Hz, 1H) 8.15-8.24 (m, 2H) 8.37 (d, J=2.23 Hz, 1H). LC-MS 256 (M+H)+.
