To a solution of 5-chloro-1-isobutyl-6-(4-methoxybenzyl)-3-methyl-1H-pyrrolo[3,4-d]pyrimidine-2,4(3H,6H)-dione (14 mg, 0.032 mmol) in CH2Cl2 (0.5 mL) is added TFA (100 μL) and TFMSA (50 μL). The mixture is stirred at r.t. for 2 h, and then carefully basified with saturated NaHCO3 aqueous solution, followed by extractions with CH2Cl2. The combined organic phase is dried over sodium sulfate, filtered, and concentrated to dryness under reduced pressure to give crude product, which is used in the next step without further purification. MS (ESI) m/z 256.1 [M+H]+.
