To a solution of 3-(dimethylamino)acrolein (0.22 mL; 218 mg; 2.2 mmol) in N,N-dimethylformamide (3 mL) was added phosphoryl chloride (0.16 mL; 269 mg; 1.76 mmol) and the mixture was stirred at room temperature for 10 min. Then N,N-diisopentyl-2-(4-methoxyphenyl)pyrazolo[1,5-a]pyrimidine-5-carboxamide (180 mg; 0.44 mmol) in N,N-dimethylformamide (2 mL) was added and the mixture was heated to 65° C. for 4 h. Then triethylamine (2 mL) and water (2 mL) were slowly added at 60° C. and the mixture was stirred for 30 min. The mixture was cooled to room temperature and diluted with methylene chloride and water. The aqueous layer was back extracted with methylene chloride and the combined organic layers were washed with water (3×10 mL), brine and concentrated. The residue was purified by flash-chromatography using 5% methanol in methylene chloride as eluant. LCMS m/z 463.3 (M+H)+, ret. time=2.98 min.
