Procedure
To a solution of 4-(tert-butoxycarbonylamino)-5-chloro-2-(2,3-difluoro-6-methoxy-benzyloxy)anisole (0.2 g) in N,N-dimethylformamide (2 mL) was added sodium hydride (55%, 21 mg), and the mixture was stirred at room temperature for 10 minutes. To the reaction mixture was added 4-chloro-3-nitropyridine (78 mg), and the mixture was stirred at room temperature overnight. The reaction mixture was poured into 0.5 mol/L hydrochloric acid, and the resulting mixture was extracted with ethyl acetate. The extract was washed with water and brine, and dried over anhydrous magnesium sulfate. The solvent was removed under reduced pressure, and the residue was dissolved in tetrahydrofuran (5 mL)—methanol (5 mL). To the solution were added nickel(II) bromide (5 mg) and sodium borohydride (53 mg) under ice-cooling, and the mixture was stirred under ice-cooling for 30 minutes, and then stirred at room temperature for 30 minutes. The reaction mixture was poured into a saturated aqueous sodium hydrogen carbonate solution, and the resulting mixture was extracted with ethyl acetate. The extract was washed with brine twice, and dried over anhydrous magnesium sulfate. The solvent was removed under reduced pressure, and the residue was purified by column chromatography on silica gel (eluent: ethyl acetate−ethyl acetate/methanol=9/1) to give 3-amino-4-{N-(tert-butoxycarbonyl)-N-[2-chloro-5-(2,3-difluoro-6-methoxybenzyloxy)-4-methoxyphenyl]amino}pyridine (53 mg). This material was dissolved in diglyme (1 mL), and the solution was stirred at 150° C. for 3 hours. The reaction mixture was poured into water, and the resulting mixture was extracted with ethyl acetate. The extract was washed with water and brine, and dried over anhydrous sodium sulfate. The solvent was removed under reduced pressure, and the residue was purified by column chromatography on silica gel (eluent: ethyl acetate/methanol=10/1). The obtained product was dissolved in ethyl acetate. To the solution were added diethyl ether and n-hexane, and the precipitated crystals were collected by filtration. The collected crystals were washed with diethyl ether, and dried under reduced pressure to give the title compound (6 mg).
