To a solution of 2-chloro-6a-methyl-5-(2-methyl-2-(tetrahydro-2H-pyran-2-yloxy)propyl)-6a,7,9,10-tetrahydro-[1,4]oxazino[3,4-h]pteridin-6(5H)-one (133 mg, 0.322 mmol) in THF (3 ml) was added 1 N HCl aqueous solution (1 ml, 1.000 mmol) and the reaction mixture was stirred for 1 h, followed by being poured into NaHCO3 aqueous solution. Extraction with EtOAc, drying over Na2SO4 and concentration in vacuo gave crude 2-chloro-5-(2-hydroxy-2-methylpropyl)-6a-methyl-6a,7,9,10-tetrahydro-[1,4]oxazino[3,4-h]pteridin-6(5H)-one (84.9 mg, 0.260 mmol, 81% yield) as a yellow oil which was used in the next step without further purification. MS [M+H] found 327.
