Procedure

4-(5-fluoro-2-methylbenzimidazol-1-yl)-3-hydroxy-Boc-piperidine (205 mg, 0.59 mmol) mmol) in THF (5 mL) and cooled to 0° C. under Ar. NaH (60% oil dispersion; 32 mg, 0.8 mmol) was added and the reaction mixture was stirred for 15 minutes. Iodomethane (0.8 mmol) was then added. After stirring for 3 hours at rt the reaction mixture was poured into saturated NaHCO3 and extracted three times with ethyl acetate. The combined organic layers were washed with brine and dried over MgSO4. Filtration followed by removal of volatiles under reduced pressure yielded crude product from which 4-(5-fluoro-2-methylbenzimidazol-1-yl)-3-methoxy-Boc-piperidine (191 mg, 89%) was isolated as a waxy solid.
