To a solution of N-(3-amino-4-methylphenyl)-3-(1-cyano-1-methylethyl)benzamide (1.00 g, 3.41 mmol) in tetrahydrofuran (15 mL) were added N-ethyl-N-isopropylpropane-2-amine (893 μL, 5.11 mmol) and 2-chloro-5-nitropyrimidin-4-yl thiocyanate (812 mg, 3.75 mmol), and the mixture was stirred at room temperature for 1.5 hr. To the reaction mixture was added saturated aqueous sodium hydrogen carbonate solution (30 mL), and the mixture was extracted with ethyl acetate (30 mL, 10 mL). The combined organic layer was washed with saturated brine (10 mL), and dried over anhydrous sodium sulfate. The insoluble material was filtered off, and the filtrate was concentrated under reduced pressure. The obtained residue was washed with ethyl acetate to give the title compound (1.41 g, 88%) as a yellow solid.
