To a RT solution of 4-chloro-7H-pyrrolo[2,3-d]pyrimidine (424 mg, 2.76 mmol) in DMF (3 mL) was added NaH (60% in mineral oil, 121 mg, 3.03 mmol). The reaction mixture was stirred for 30 min at RT and then a solution of methanesulfonic acid 3-chloro-1-phenyl-propyl ester in DMF (2 mL) was added at 0° C. The resulting mixture was stirred overnight at RT and then diluted with H2O and EtOAc. The organic layer was separated, washed 3 times with water, dried over MgSO4, filtered, concentrated, and purified via flash chromatography (hexane/EtOAc), affording 4-Chloro-7-(3-chloro-1-phenyl-propyl)-7H-pyrrolo[2,3-d]pyrimidine as a colorless oil (480 mg, 56% yield).
