Procedure
39 mg of the 4-(8-acetylimidazo[1,2-a]pyridin-3-yl)-2-{[(1S)-1-(4-piperazin-1-ylphenyl)ethyl]amino}pyrimidine-5-carbonitrile [156-3] was dissolved in a mixture solvent of 1 mL of tetrahydrofuran and 1 mL of methanol, then 2.8 mg of sodium boronhydride was added thereto, and stirred at room temperature for 15 minutes. Thereto, a saturated aqueous solution of sodium hydrogen carbonate was added, and the mixture was extracted with a mixture solvent of chloroform and methanol (chloroform:methanol=9:1). The organic layer was washed with saturated brine and dried over anhydrous sodium sulfate. The insolubles were filtered, and then the filtrate was concentrated under reduced pressure. The obtained residue was purified by preparative thin-layer chromatography, and 31.9 mg of 4-[8-(1-hydroxyethyl)imidazo[1,2-a]pyridin-3-yl]-2-{[(1S)-1-(4-piperazin-1-ylphenyl)ethyl]amino}pyrimidine-5-carbonitrile [156-4] was obtained as a two kinds of diastereomer mixture.
