Procedure
The product from step A was dissolved in THF (10 mL) then CDI (1.018 g, 6.28 mmol) was added. After being stirred for one hour, β-alanine ethyl ester hydrochloride (560 mg, 6.28 mmol) was added, then the mixture was stirred overnight at RT. The mixture was diluted with EtOAc then washed with water then brine. The organic layer was dried over MgSO4, filtered, then concentrated. The resulting residue was purified by silica gel chromatography to afford the title compound. 1H NMR (500 MHz, CD3OD): δ 7.72 (d, J=2.5 Hz, 1H); 7.67 (d, J=8.5 Hz, 2H); 7.50 (dd, J=9.0, 2.5 Hz, 1H); 7.37 (d, J=8.0 Hz, 2H); 7.24 (d, J=8.0 Hz, 2H); 7.13 (d, J=8.5 Hz, 2H); 6.81 (d, J=9.5 Hz, 1H); 5.63 (d, J=6.0 Hz, 1H); 4.12 (q, J=7.5 Hz, 2H); 3.61 (t, J=7.0 Hz, 2H); 3.19 (m, 1H); 2.62 (t, J=7.0 Hz, 2H); 2.00-1.94 (m, 1H); 1.73-1.67 (m, 1H); 1.22 (t, J=7.5 Hz, 3H); 1.19-1.16 (m, 2H); 0.83 (t, J=7.0 Hz, 3H). LC2 2.38 min. (M+H)+ 597.
