Procedure

To a suspension of 6-(6-methoxy-2-pyrimidin-5-yl-1H-indol-3-ylmethyl)pyridine-2-carboxamide (351 mg) in N,N-dimethylformamide (9.8 mL) was added phosphoryl chloride (0.133 mL) under ice-cooling, and the mixture was stirred under ice-cooling for 3 hours. To the reaction mixture were added a saturated aqueous sodium hydrogen carbonate and water, followed by extraction with ethyl acetate. The organic layer was washed with water and saturated brine successively, dried over anhydrous sodium sulfate, and then concentrated under reduced pressure. The residue was purified by aminopropylated silica gel column chromatography (eluting solvent: methanol-ethyl acetate) to obtain the title compound (326 mg). 1H-NMR (CDCl3) δ ppm: 3.87 (3H, s), 4.40 (2H, s), 6.81 (1H, dd, J=2.3, 8.5 Hz), 6.90-6.95 (1H, m), 7.30-7.39 (2H, m), 7.50-7.57 (1H, m), 7.64-7.72 (1H, m), 8.20 (1H, br s), 9.00 (2H, s), 9.20 (1H, s).
