7-Methyl-1-oxo-2-(4-trifluoromethoxy-benzyl)-2,3-dihydro-1H-isoindole-5-carboxylic acid (35 mg, 0.096 mmol), 1-methylpiperazine (12 μL, 0.11 mmol), EDCI (20 mg, 0.11 mmol), HOBT (14 mg, 0.11 mmol) were stirred in DMF (5 mL) for 18 hours at room temperature. The reaction mixture was partitioned between EtOAc and water, and the organic layer was washed with brine, dried over anhydrous sodium sulphate, filtered and concentrated to yield 25 mg of a brown solid (70%). 1H NMR (300 MHz, CDCl3): δ 7.34-7.37 (m, 2H), 7.19-7.24 (m, 4H), 4.79 (s, 2H), 4.24 (s, 2H), 3.84 (br s, 2H), 3.44 (br s, 2H), 2.79 (s, 3H), 2.55 (br s, 2H), 2.36 (s, 5H).
