To a solution of 5-biphenyl-4-yl-6-chloro-2-(methylsulfonyl)-1H-benzimidazole (5.37 g, 14.0 mmol) in THF (70 mL) was added DIPEA (3.67 mL, 21.0 mmol) followed by SEMCl (3.7 mL, 21.0 mmol). The reaction was maintained for 5 h at ambient temperature. Then the reaction mixture was diluted with EtOAc and washed with saturated aqueous NaHCO3, followed by brine. The organic phase was dried over Na2SO4, filtered, and concentrated. Chromatography of the resulting residue over silica eluting with 5-25% EtOAc/hexanes afforded the desired product as a white solid. LC-MS: calculated for C14H20ClN2O3SSi 485.97, observed m/e 428.83 (M+H)+ (Rt 2.30 min).
