Procedure
To the solution of (S)-2-methyl-6,7-dihydro-5H-[1]pyrindin-5-ylamine hydrochloride (75 mg, 0.41 mmol) and (3-cyclopropyl-4-difluoromethyl-1-methyl-1H-pyrazolo[3,4-b]pyridin-6-yloxy)-acetic acid (building block A2, 110 mg, 0.37 mmol) in DMF (6 ml) were added DIPEA (0.26 ml, 1.48 mmol) and HATU (169 mg, 0.44 mmol) and the reaction was stirred at it for 3 h. The mixture was diluted with EtOAc, washed with aqueous bicarbonate solution and brine, dried over sodium sulfate, and the solvents were evaporated under reduced pressure. Purification by preparative HPLC (Gilson GX-281, Waters Sunfire C18, 5 μm, 30×100 mm with guard column 19×10 mm, solvent A: water with 0.1% TFA, solvent B: acetonitrile, gradient 10-30% B in 16 min, flow rate 50 ml/min) gave the title compound as off-white solid (115 mg, 72%). [1H-NMR (DMSO-d6, 600 MHz) δ 8.61 (d, 1H), 7.47 (t, 1H), 7.36 (d, 1H), 6.99 (d, 1H), 6.93 (s, 1H), 5.32 (dd, 1H), 4.89 (s, 2H), 3.81 (s, 3H), 3.34 (s, 3H), 2.93-2.89 (m, 1H), 2.86-2.80 (m, 1H), 2.41 (s, 3H), 2.41-2.36 (m, 1H), 2.24-2.19 (m, 1H), 1.92-1.86 (m, 1H), 0.94-0.87 (m, 4H); LCMS RtI=3.032 min; [M+H]+=428.2]
