A mixture of rac-(4-{[(2′S,3′R,4′S,5′R)-6-chloro-4′-(3-chloro-2-fluoro-phenyl)-2′-(2,2-dimethyl-propyl)-2-oxo-1,2-dihydro-spiro[indole-3,3′-pyrrolidine]-5′-carbonyl]amino}-phenoxy)-acetic acid prepared in Example 206 (23 mg, 0.037 mmol), N,N-diisopropylethylamine (33 mg, 0.258 mmol) and O-(7-azabenzotriazol-1-yl)-N,N,N′,N′-tetramethyluronium hexafluorophosphate (HATU) (Chem-Impex, 24 mg, 0.062 mmol) in DMF (4 mL) was stirred for 20 min before NH4Cl (6 mg, 0.099 mmol) was added. The mixture was stirred for 0.5 h and diluted with EtOAc (70 mL), washed with water (10 mL), brine (15 mL) and concentrated. The crude product was purified by flash chromatography (EtOAc/hexane, 25/75 to 95/5) to give rac-(2′S,3′R,4′S,5′R)-6-chloro-4′-(3-chloro-2-fluoro-phenyl)-2′-(2,2-dimethyl-propyl)-2-oxo-1,2-dihydro-spiro[indole-3,3′-pyrrolidine]-5′-carboxylic acid (4-carbamoylmethoxy-phenyl)-amide as a white solid (9 mg, 40%).
