20 g (129 mmol) of 6-amino-1,3-dimethyluracil was suspended in 550 ml of dimethylformamide. 216 g of phosphorus oxychloride was added to the suspension at a temperature below 20° C., and the reaction mixture was stirred for 30 minutes. The resulting precipitate was filtered off and washed with acetone to give 6-imino-1,3-dimethyl-5-dimethylaminomethylene-5,6-dihydrouracil hydrochloride (compound 1) in a 95% yield. The product may also be used in the following reaction without further purification.
