To a solution of trans-4-(2,4-Dioxo-1,4-dihydro-2H-quinazolin-3-ylmethyl)-cyclohexanecarboxylic acid (1.5 g, 4.96 mmol) in dimethylacetamide (10 mL), HATU (1.88 g, 4.96 mmol) and triethylamine (0.76 mL, 5.45 mmol) were added and the mixture left stirring at room temperature for 1 hour. Then tert-butyl-1-piperazinecarboxylate (0.92 g, 4.96 mmol) was added and the reaction left stirring for 18 h at room temperature. The solvent was removed under reduced pressure and the residue was washed with a 0.4M Na2CO3 solution. The titled compound was obtained as a white solid (2.3 g, quantitative yield).
