To a solution of step A product, 3-2-a, (0.85 g, 2.4 mmol) in DMF (15 mL), was added NCS (0.42 g, 3.15 mmol) at room temperature. The reaction mixture was heated at 60° C. for 1 h. The reaction mixture was cooled to 0° C. and ethyl propiolate (0.67 g, 6.8 mmol) was added. Then Et3N (0.3 g, 3 mmol) in DMF (5 mL) was added dropwise over 20 min. The reaction mixture was slowly allowed to warm to room temperature and stirred for overnight. The mixture was diluted with water (50 mL) and extracted with EA (100 mL). The organic layer was washed with brine (50 mL), dried over Na2SO4, and concentrated in vacuum. The residue was purified by column chromatography with the eluent (PE:EA=2:1) to afford 3-2 (150 mg, 14%) as a light yellow solid. 1H NMR (300 MHz, DMSO-d6) δ10.04 (s, 1H), 8.9 (s, 1H), 7.68 (s, 1H), 7.35-7.26 (m, 11H), 5.76 (s, 1H), 4.40 (q, J=7.2 Hz, 2H), 1.34 (t, J=7.2 Hz, 3H). LC-MS: (M+H)+ 445.
