To a solution of 5-amino-2-chloroisonicotinic acid (5 g, 0.029 mol) in DMF (40 mL) was added EDCI (8.34 g, 0.0435 mol) and HOBT (5.87 g, 0.0435 mol). The reaction was stirred at RT for 30 min then 3-(tert-butyldimethylsilyloxy)propan-1-amine (5.5 g, 0.029 mol) was added. The reaction was stirred at RT for 5 h then poured in EA (50 mL) and water (20 mL). The organic layer was washed with brine (50 mL), dried over Na2SO4 and concentrated to a residue which was purified by chromatography eluted with PE/EA (2:1) to give 5-amino-N-(3-(tert-butyldimethylsilyloxy)propyl)-2-chloroisonicotinamide (4.3 g, 43% yield) as a white solid. LCMS: MH+ 344 and TR=1.877 min.
