To a mixture of (R)-piperidin-3-yl(pyrrolidin-1-yl)methanone in N,N-dimethylformamide (120 mL) was added diisopropylethylamine (15 mL) followed by 6-chloro-3-nitropyridin-2-amine (3.47 g, 20 mmol). The mixture was stirred at 50° C. for 16 h. The solvent was removed under reduced pressure. The residue was purified via flash column chromatography over silica gel (ethyl acetate:petroleum ether, 1:10 to 10:1) to afford (R)-(1-(6-amino-5-nitropyridin-2-yl)piperidine-3-yl)(pyrrolidin-1-yl)methanone (4.838 g, 76.3%).
