Procedure

To a solution of 4-[4-(cyclopropylsulfonyl)phenyl]-5-[(2R,3R,7R)-2,3-diphenyl-1,4-dioxaspiro[4.4]non-7-yl]pent-1-en-3-one (500 mg) in ethanol (5 mL) were added pyridine-2-carbaldehyde (107 μL), 3-benzyl-5-(2-hydroxyethyl)-4-methyl-1,3-thiazol-3-ium chloride (28 mg) and triethylamine (56 μL), and the mixture was stirred with heating under reflux for 2 hr. After cooling to room temperature, the reaction mixture was diluted with ethyl acetate and washed with water. The ethyl acetate layer was washed with saturated brine, dried (MgSO4) and concentrated to give a crude product (610 mg). To a solution of the obtained crude product (610 mg) in acetic acid (5 mL) was added ammonium acetate (1135 mg), and the mixture was stirred at 110° C. for 45 min. After cooling to room temperature, the reaction mixture was diluted with ethyl acetate and washed with water. The ethyl acetate layer was washed with saturated aqueous sodium hydrogen carbonate and saturated brine, dried (MgSO4) and concentrated. The residue was subjected to preparative HPLC to give the title compound (70 mg, yield 12%) as a colorless amorphous solid. MS: 631 (MH+).
