In an argon atmosphere, TrCl (936 mg, 3 mmol) and Et3N (556 μL, 3 mmol) were added to a solution of 1H-tetrazole-5-acetic acid (128 mg, 1 mmol) in THF (10 mL) and stirred for 3 hours at room temperature. The reaction mixture was diluted with EtOAc, and the organic layer was washed with water and dried over anhydrous sodium sulfate. The solution was filtered, and the filtrate was concentrated at a reduced pressure. The resulting residue was purified by neutral flash silica gel column chromatography (1.5×6, MeOH in CH2Cl2 0-0.5%) to give 2-(1-trityl-1H-tetrazol-5-yl)acetic acid (62 mg, 18%) as a white solid.
