Racemic 2-hydroxy-2-methyl-3-[4-(3-methyl-5-{[4-(trifluoromethyl)pyrimidin-2-yl]amino}phenyl)-1H-pyrazol-1-yl]propanoic acid (142 mg, 0.26 mmol), HOBT (61 mg, 0.4 mmol), EDC (76 mg, 0.4 mmol), and ammonium chloride (42.5 mg, 0.8 mmol) were combined with DMF (2.5 mL) in a flask. Triethylamine (0.35 mL, 2.51 mmol) was added and the mixture was stirred for 3 days at room temperature. The mixture was diluted with EtOAc and diluted with 1:1 water:brine (3×). The organic layer was dried over sodium sulfate, filtered and concentrated under reduced pressure to afford racemic 2-hydroxy-2-methyl-3-[4-(3-methyl-5-{[4-(trifluoromethyl)pyrimidin-2-yl]amino}phenyl)-1H-pyrazol-1-yl]propanamide. MS ESI calcd. for C19H19F3N6O2 [M+H]+ 421. found 421.
