Triphosgene (0.281 g, 0.948 mmol) (Sigma-Aldrich) in THF (5 mL) was added to a to a solution of 7-amino-3,4-dihydroquinolin-2(1H)-one (0.4659 g, 2.87 mmol) (Astatech, Inc) and DIEA (2.01 mL, 11.5 mmol) (Aldrich) in THF (28 mL) at room temperature. After 10 minutes (R)-7,8-difluoro-2,2-dimethylchroman-4-aminium chloride (0.717 g, 2.87 mmol) (B-8; WO2010/04401) was added and the mixture was stirred for an additional 2 hours. The mixture was diluted with saturated aqueous NaHCO3 (30 mL) and extracted with EtOAc (3×30 mL). The combined organic extracts were washed with 1 N aqueous HCl (30 mL) and concentrated. The residue was purified by silica gel chromatography (5% MeOH/CH2Cl2) to afford the title compound (0.970 g, 2.42 mmol, 84% yield). 1H NMR (300 MHz, DMSO-d6) δ 10.01 (s, 1H), 8.50 (s, 1H), 7.14-6.84 (m, 5H), 6.44 (d, J=8.6 Hz, 1H), 4.95 (dd, J=15.7, 9.7 Hz, 1H), 2.78 (t, J=7.5 Hz, 2H), 2.41 (dd, J=8.4, 6.5 Hz, 2H), 2.16 (dd, J=13.4, 6.1 Hz, 1H), 1.80 (dd, J=13.2, 11.3 Hz, 1H), 1.45 (s, 3H), 1.32 (s, 3H); MS (ESI+) m/z 402 (M+H)+, 400 (M−H)+.
