Procedure
To a solution of 1-(tert-butoxycarbonyl)-5,5-difluoropiperidine-3-carboxylic acid (2.1 g, 7.92 mmol) in N,N-dimethylformamide (100 mL) were added 1,1′-carbonyldiimidazole (CDI, 2.56 g, 15.82 mmol), dimethylamine hydrochloride (1.29 g, 15.82 mmol) and triethylamine (2.4 g, 23.71 mmol). After 16 h, the mixture was diluted with ethyl acetate, then was washed sequentially with cold water (×2) and brine, The organic layer was dried over sodium sulfate, filtered, and concentrated under reduced pressure. The resulting residue was purified by silica gel chromatography to afford tert-butyl 5-(dimethylcarbamoyl)-3,3-difluoropiperidine-1-carboxylate (1.4 g, 60% over two steps) as a pale yellow solid.
